Workshop on Genome Editing using CRISPR-Cas9

The aim of this workshop is to provide hands-on training in genome editing in mammalian cells using the powerful CRISPR/Cas9 system. It will cover topics from sg RNA design, molecular cloning and transfection, through off-target assessment, validation and different applications.

 

Target Audience:

This workshop is directed for Researcher assistants, PhD students, Post-doc fellows and technicians with a molecular biology background who want to get familiar with genome editing in mammalian cells using CRISPR/Cas9 system or who are already using CRISPR but would like to receive focused training to optimize their experiments. Although no previous experience in genome engineering is required, basic skills on molecular and cell biology will be an advantage.

CONTENTS

 

Theory concepts:

• Nuclease-induced genome editing: NHEJ vs HDR

• Overview of CRISPR-Cas9 systems: nuclease, nickase and catalytic-dead

• Applications of CRISPR-Cas9 for DNA editing (knock-in and knock-out), epigenome editing, gene regulation and imaging

• Generation of in-vivo models

• High-throughput genetic screens

 

Practical topics:

• Bioinformatic design of guide RNAs and donor templates

• Cloning of guide RNAs into expression vectors

• Transfection of a cell line with Cas9 plasmids and guide RNAs

• Accessing the efficiency of DNA editing

• Specificity of CRISPR reagents (off-target effects, and how to deal with them)

• Validation of clonal cells edited by Cas9
